     A+C線維刺激---0.5ms pulse duration; 4Hz at 200xTH (>2mAmp)
  • CMA/20 microsialysis probe (CMA/Microdialysis, Acton, MA; 20kDa mol, wt cutoff, 4mm probe legth)
     Using infusion pump lactated ringers solution (pH7.4) was infused (3μl/min) for a 120 min washout period, followed by a 60 min collection period to establish EAA sampling every 10 min.
      Samples were analyzed for EAA concentrations using reverse phase high performance liquid chromatography (HPLC) equipped with a fluorescence detector.

  • 浮腫(thickness)、発赤(score)、皮膚温(contact thermode):CFAの注入前、2,7,14日後の左右差を比較。どれも2日目が一番変化あり。
  • 電顕:digital nervesの無髄軸索の NMDAR1は normal ratにもあるが、後肢足蹠CFA注入2,7日後に増加しているが、14日ではnormalと有意差なし。
  • 行動:(1)正常ラットでは、足蹠への1 mM NMDA注入で、von Freyでの機械刺激による感受性が有意に閾値低下。
     (2)CFA炎症ラットの機械刺激による感受性の閾値は正常ラットよりも低く、CFA炎症ラットへでは、0.5 mM NMDA注入でも有意に閾値低下。
     (3)CFA注入2日後に同側足蹠にMK-801を注入すると、閾値の低下から回復(PBS注入,対側足蹠に注入したMK-801と比較)。
  • 電気生理:In vitro skin-nerve: medial and lateral plantar nerves, single-unit studies, C-mechanoheat (CMH) receptors or C-mechanoreceptors (CM)
    (1)2-day CFA-inflamed glabrous skinでは、熱刺激の閾値の低下
    (2)足蹠に投与されるNMDAによって興奮する受容器の割合および発火数の増加はinflamed skinのほうが多く、dose dependencyがある。
    (3)NMDA2回注入により感受性が亢進。2回目にMK-801も一緒に注入すると、健常皮膚では初回注入時と同じ反応性、炎症皮膚では初回注入時よりもスパイク数減少。
    (3) NMDA注入前後で、熱反応を比較。健常皮膚では、NMDA注入後に閾値の低下と発火数増加。炎症皮膚では、閾値の低下のみ。
  • 末梢NMDA受容体の活性化は急性の侵害受容と持続性の炎症における痛みに関与する。炎症動物時での、末梢軸索にNMDAR1のアップレギュレーション(?)と活性化が、持続する炎症に伴われる機械痛覚過敏と熱痛覚過敏に関与する。鎮痛薬の末梢でのターゲットとなる可能性がある。
